Reduction of [Ca2+]i from 1.8 to 0.2 mM reduced ATP- and TPA-activated currents by 51 and 53%, respectively. Intracellular application of the purified protein kinase C (PKC) inhibitor, 1-(5-isoquinolinyl-sulphonyl)-2-methylpiperazine hydrochloride (H7), reduced the current activated by TPA, but not the ATP-activated current, when it was applied before the application of ATP. The synergistic reduction of ATP-activated currents was observed when H7 was applied immediately after the activation of the channel with ATP.
