Carriers of the PD-1 Th1 haplotype (C) displayed higher osteoclastogenesis compared with noncarriers (T). Knockdown of PD-1 by RNA interference reversed the increased osteoclastogenesis. Osteoclastogenesis was associated with an increase in macrophages expressing CD226, a marker for macrophage migration inhibitory factor receptor, and was independent of PD-1 and inducible co-stimulator 1 and CTLA-4.
